Determination of PML/RARa Gene Rearrangement In Acute Promyelocytic Leukemia with Reverse Transcription Polymerase Chain Reaction

Abstract
The Molecular characterization of the translocations involved in hematopoietic tumors has advanced our understanding of neoplastic transformation. Acute promyelocytic leukemia(APL:AML M3) is identified by a unique t(15;17) translocation which fuses the PML gene to the retinoic acid receptor alpha gene(RAR¥á) and a consistent translocation is present in the blasts of the majority of APL patients. The t(15;17) translocation generates a PML/RAR¥á chimeric gene that is transcribed as a fusion PML/RAR¥á messenger RNA. The detection of the PML/RAR¥á rearrangements involved in APL has not only provided pathogenitic marker but also permitted more accurate diagnosis and monitoring of the patient¡¯s response to therapy. I had examined the PML/RAR¥á rearrangement by reverse transcriptase polymerase chain reaction (RT-PCR). In APL patients, I had detected long form type(second PCR product 370 bp) and short form type(first PCT product 221 bp) gene rearrangement.
